Western blotting: 1:500
The optimal dilution for a specific application
should be determined by the researcher.
Example of Application
Poly-ubiquitination of lysozyme
1. Prepare reaction mixture as follows:
10 μl 1M Tris-HCL (pH 9.0)
1.8 μl 300 mM MgCl2
1.8 μl 200 mM ATP
0.6 μl 100 mM DTT
1 μl 200 U/μl Inorganic Pyrophosphatase
45 μl E1, E2, E3 Mixture (Cat. No. BC-066)
15 μl 2 mg/ml Lysozyme
15 μl 10 mg/ml Ubiquitin
9.8 μl Deionized Water
Total 100 μl
2. Incubate the reaction mixture at 37oC for 90
minutes.
Western blot
3. Run 5 μl/lane of the above reaction mixture
in SDS-PAGE (12.5).
4. Transfer proteins to a membrane at 6V for
90 min.
5. Visualize the bands of poly-ubiquitinated
lysozyme with anti-lysozyme antibody at a
dilution of 1:500 followed by alkaline
phosphatase-conjugated rabbit anti-mouse
IgG. Color development was done with
NBT/BCIP.
