Product: Anti-ADAM10 (C-terminus) pAb
Cat. No.: PC-047 (100 μg)
Background:
Proinflammatory cytokine tumor necrosis factor-
a (TNF-a) contributes to a variety of
inflammatory responses and programmed cell
death. Notch receptor and its ligand participate
in cell fate decisions during vertebrate
development and are associated with several
human disorders, including a T-cell lymphoma.
TNF-a, notch and its ligand delta are all
membrane-bond molecules, which are cleaved
by proteases to release mature proteins or
functional receptor. ADAM10, a metalloprotease-
disintegrin in the family of mammalian
ADAM (for a disintegrin and metalloprotease),
was recently identified to cleave TNF-a, notch
and its ligand delta. The genes encoding human,
mouse, and bovine ADAM10 were recently
cloned and designated ADAM 10, kuzbanian
(KUZ), and MADM, respectively. ADAM10
mRNA is expressed in a variety of human and
bovine tissues.
Specificity:
This antibody recognizes ADAM10.
Species Reactivity:
Human, mouse, and rat. Other species not
tested.
Host:
Rabbit
Ig Isotype:
IgG
Immunogen:
A peptide corresponding to amino acids 732 to
748 of human ADAM10. This sequence is
identical to those of bovine and rat origins and
differs from that of mouse ADAM10 by one
amino acid.
Format:
200 μL at 500 μg/mL polyclonal antibody in PBS
containing 0.02% sodium azide. Purified by ion
exchange chromatography.
Storage:
Store at 4°C.
Applications and Suggested Dilutions:
Immunohistochemistry: Use at 2 μg/mL.
Western blot: Use at a 1:500-1:2,000 dilution.
The optimal dilution for a specific application
should be determined by the researcher.
Positive Control:
Jurkat whole cell lysate. An approximately 85
kDa band, which may represent precursor, can
be detected.

Limitations:
For in vitro research use only. Not for use in
diagnostics or in humans.
